Ultrastructural characterization of mouse embryonic stem cell-derived oocytes and granulosa cells.

چکیده
Germ cells are a unique population of cells responsible for transmitting genetic information from one generation to the next. Our understanding of the key mechanisms underlying germ cell development in vivo remains scarce because of insufficient amounts of cell materials available for conducting biological studies. The establishment of in vitro differentiation models that support the generation of germ cells from mouse pluripotent stem cells provides an alternative means for studying reproductive development. The detection and analysis of stem cell-derived germ cells, however, present technical challenges. Methods for determining the developmental stage of germ cells ex vivo, such as gene expression and/or immunochemical analyses are inadequate, frequently necessitating the use of alternative, elaborate methods to prove germ cell identity. We have generated putative oocytes and granulosa cells in vitro from mouse embryonic stem cells and utilized electron microscopy to characterize these cells. Here, we report on the striking ultrastructural similarity of in vitro-generated oocytes and granulosa cells to in vivo oocytes developing within follicles.

The Effect of Cardio Gel and Matrigel on the Ultrastructure of Cardiomyocytes Derived From Mouse Embryonic Stem Cells
Purpose: To investigate the effect of cardiogel and matrigel on the ultrastructure of embryonic stem cell-derived cardiomyocytes. ECM: Extracellular Matrix derived from cardiac fibroblasts (cardiogel), commercial extracellular matrix (matrigel) and control group (without ECM) were cultured for up to 21 days. Ultrastructural properties of cardiomyocytes were evaluated by transmitting electron mi...
متن کاملEffect of Different Concentrations of Forskolin Along with Mature Granulosa Cell Co-Culturing on Mouse Embryonic Stem Cell Differentiation into Germ-Like Cells
Background: Germ cell development processes are influenced by soluble factors and intercellular signaling events between them and the neighboring somatic cells. More insight into the molecular biology of the germ cell development from embryonic stem (ES) cells and investigation of appropriate factors, specifically those targeting differentiation process, is of great importance. In this study, w...
متن کاملExtract of mouse embryonic stem cells induces the expression of pluripotency genes in human adipose tissue-derived stem cells
Objective(s): In some previous studies, the extract of embryonic carcinoma cells (ECCs) and embryonic stem cells (ESCs) have been used to reprogram somatic cells to more dedifferentiated state. The aim of this study was to investigate the effect of mouse ESCs extract on the expression of some pluripotency markers in human adipose tissue-derived stem cells (ADSCs). Materials and Methods: Human A...

Purpose: Differentiation of mouse embryonic stem cells into Insulin secreting endocrine cells. Materials and Methods: In this study, Royan B1 mouse embryonic stem cell (derived from C57BL/6 mouse) were used. In directed differentiation method, embryonicstem cells after embryoid bodies formation were differentiated into insulin secreting cells. Nestin positive cells were obtained after culture ...
متن کاملEmbryonic stem cells derived cardiomyocytes are a suitable model for assessment of cardiotoxic effects of doxorubicin and other drugs
Introduction: Doxorubicin is frequently used for treatment of several types of cancer. Doxorubicin cardiac toxicity has limited the use of this drug. Corticosteroids may prevent doxorubicin induced cardiotoxicity. Therefore the aim of this study was to evaluate mouse embryonic stem cells derived cardiomyocytes as a model to evaluate the effect of Doxorubicin and dexamethasone. Methods: Mouse ...
